Mechanism of Action

Ponatinib (Iclusig) is a third-generation pan-mutational BCR-ABL tyrosine kinase inhibitor whose ethynyl linker enables binding to the T315I-mutated BCR-ABL kinase domain, inhibiting proliferation driven by all known BCR-ABL point mutations—including T315I—in CML and Ph+ ALL.
