Mechanism of Action
Lorlatinib competitively inhibits the kinase activity of ALK/ROS1 through a large ring structure, blocking downstream carcinogenic pathways such as PI3K/AKT and RAS/MAPK. At the same time, it overcomes drug resistance mutations such as G1202R and efficiently penetrates the blood-brain barrier. It is currently the third-generation ALK inhibitor with the broadest resistance spectrum and the strongest efficacy in treating brain metastases.
